Effect of IGF-¥± against methylmercury-induced neurotoxicity in the cultured mouse cerebral cortical neurons

Abstract
In order to examine the effect of insulin-like growth factor-¥±(IGF-¥±) on the cultured mouse cerebral cortical neurons inhibited by the neurotoxicity induced by methylmercuric chloride(MMC), MTT assay and neurofilament enzymeimmunoassay were performed after cerebral cortical neurons were preincubated with various concentrations of IGF-¥± before treatment of cells with MMC. The results were as follows. 1) MMC induced cell degeneration such as the decrease of cells in number and the loss of neurites in the cultured mouse cerebral cortical neurons. 2) MTT50 value was a 20 uM MMC. 3) IGF-¥± was effective in the increase of cell viability of the neurons inhibited by MMC and in neurofilamental differentiation in a dose dependent manner. 4) MMC was toxic on the cultured neurons in a time-and dose-dependent manners. From the above results, it is suggested that methylmercury induces the inhibition of cell viability in cultured mouse cerebral cortical neurons and selective neurotrophic factors such as IGF-¥± are effective in cultured neurons inhibited by methymercury.
